- Researchers can use multiple sgRNA guides targeting the same genomic locus to create deletions larger than those that arise from a single guide experiment. ICE’s new Multiplex Analysis enables you to analyze those experiments by allowing for up to three guide sequences for one sample The algorithm will consider all possible pairwise multiplex edits and return the ICE editing efficiency and KO score. The webtool will display the multiplex editing results in the same user-friendly format, with additional information elements to highlight the presence of multiplex edits.
Knockout Score: The Knockout or KO Score assumes that the target of interest lies within a coding sequence and calculates the percentage of frameshift indels that fall out of frame from your intended edit and indels that are large enough (21+bp) to be likely to affect protein function. This score is useful for determining if your gene editing experiment has resulted in efficient knockout of protein function. This replaces the ICE-D score in all results.
We renamed the Discord and Indel tab to Indel Distribution, and re-organized the results to place the Indel Plot in the primary position within the graph, and reorganized the position of our Detail results tabs: The contributions tab now appears first, followed by Indel Distribution and finally Trace data.Based on customer feedback since the v1.0 release, we have removed the Alignment tab.
