To Knockin a sequence into the genome, the double-stranded break induced by Cas9 nuclease should be followed by homology-directed repair (HDR). The cell needs a homologous DNA template with the desired final genomic sequence, along with the target sgRNA and Cas9, for the HDR process to be effective. Only a relatively small percentage of the cells actively use this DNA template to repair the double strand break via homologous recombination, thereby incorporating the desired sequences into the genome.
