The Guide Verification Tool helps you fully understand all the relevant characteristics of any guide RNA, enabling you to make informed experimental decisions regarding the use of a guide sequence. Once you input a guide RNA sequence from any design source, including Synthego's CRISPR Design Tool, the Verification Tool will show you the PAM site and cut site along with the DNA strand that the guide RNA will bind.
Importantly, this tool will also display detailed information regarding the target gene, whether the guide RNA targets an early region in the gene, ability to target a common exon, on-target activity score and whether it has any locations with zero, one or two mismatches. The tool also displays detailed information on the locations of the potential off-target sites, including the number and location of the mismatches. Together, this information allows you to assess the quality of your guide RNA, including its ability to effectively and specifically cut at a site in the genome.
You can access the verification tool directly through the following link: https://design.synthego.com/#/validate
Simply enter the target genome, and the guide sequence then click validate to view the information described above.
You can also access the verification tool by clicking on any guide sequence found through the design tool to open a new window with the verification information.
